The selective biocatalytic synthesis and investigation of the crystal structure of (2R,6R)-hydroxyketaminium chloride were successfully performed by Joost Lülf and Ansgar Bokel from our institute in cooperation with Guido Reiß from the Institute of Inorganic Chemistry and Structural Chemistry, HHU.

“(2R,6R)-hydroxyketamine is one of the minor human metabolites of the anesthetic drug (R,S)-ketamine. Whereas the hepatic metabolism by cytochrome P450s (CYPs) leads to a mixture of several hydroxynorketamines and hydroxyketamines, mutagenesis of the bacterial CYP154E1 allowed the selective synthesis of (2S,6S)-hydroxynorketamine from (S)-ketamine as well as (2R,6R)-hydroxynorketamine from (R)-ketamine. In this study, the crystal structure of one of the side products of the latter reaction is presented.”
